vagrant@lucid32:~$ sudo passwd -u root
passwd: password expiry information changed.
Then change the root password:

Retype new UNIX password:

Default Root Password? - Ubuntu Forums
The root password at fresh install of Ubuntu (10.04) at least is not set. Set it by sudo passwd root and follow instructions
